Antioxidant and Anti-Inflammatory Effects of Abies holophylla Leaf Extract in LPS-Induced RAW 264.7 Cells

|
Abies holophylla is an evergreen and coniferous tree that is widely distributed in Russia, China, and Korea. This study investigated the antioxidant, antidiabetic, and anti-inflammatory effects of Abies holophylla leaf ethanol extract (AHLE). The total polyphenol content of the AHLE was 188.09 mg tannic acid equivalent/mg. 1,1-Diphenyl-2-picrylhydrazyl radical scavenging activity of AHLE were remarkably increased in a dose-dependent manner (P<0.05), and was about 89.63% at 1 mg/mL concentration. Superoxide dismutase activity of AHLE was 95.61% at 1 mg/mL concentration. ¥á-Glucosidase inhibitory activity of AHLE was 51.79% at 1 mg/mL concentration and was increased in a dose-dependent manner. Nitrite scavenging activity of AHLE was 84.73%, 66.31%, and 42.40% at pH of 1.2, 3.0, and 6.0 at 1 mg/mL concentration, respectively. The reactive oxygen species level was examined using lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. The ROS level of cells was reduced by 15.94% with AHLE at 50 ¥ìg/mL, as compared with the LPS-treated group. The anti-inflammatory effects were examined using LPS-stimulated RAW 264.7 cells. Nitric oxide production was reduced 31.10% by the addition of AHLE at 50 ¥ìg/mL. AHLE reduced the protein expression of inducible nitric oxide synthase in a dose-dependent manner (P<0.05). From these results, we conclude that Abies holophylla leaf may be a highly valuable natural product owing to its high quality functional components, as well as its antioxidant, antidiabetic, and anti-inflammatory effects.
